摘要

Abstract

Objective To investigate the regulation of methyl ferulic acid on the growth, movement and interstitial transition of glioma cell U251 by inhibiting the activation of P38.Methods The glioma cell line U251 was selected as the study object.The drug-administered group was trea-ted with 25, 50, 100 μM methyl ferulic acid for 48 h, the cell proliferation was detected by the EdU cell proliferation assay kit, the cell invasion was detected by TRANSWELL, the cell migration was detected by scratch test, the expressions of Ki67, PCNA, N-cadherin, E-cadherin, p-P38, p-MAPKAPK2 and p-HSP27 were detected by Western blotting.The pc-P38 MAPK plasmid was transfected with 100 μmol/L methyl ferulic acid for 48 h alone or in combination, and the above in-dicators were again detected.Results Compared with the Control group, the proportion of EdU-positive cells in the methyl ferulic acid treatment group was significantly reduced, the expression of Ki67 and PCNA was significantly down-regulated, the number of invasive cells was significantly re-duced, the scratches were broadened, the healing rate was significantly reduced, the expression of E-cadherin was significantly up-regulated, the expression of N-cadherin was down-regulated, the cell morphology was significantly improved, and the expression of p-P38, p-MAPKAPK2 and p-HSP27 was significantly down-regulated, and the effect was enhanced with the increase of the dose ( P<0.01) ; Co-transfection of pc-P38 MAPK plasmid reversed the regulation of methyl ferulic acid inhibition of P38 activation on growth, migration and mesenchymal transition of glioma cell line U251.Conclusion Methyl ferulic acid inhibits the growth, movement and epithelial-mesenchymal transition of glioma U251 cells by inhibiting the activation of P38.
