Assessing the causal effect of vitamin D levels on the risk of atrial fibrillation: a Mendelian randomization study

Abstract
Objective: This study aimed to determine whether vitamin D levels are causally associated with atrial fibrillation (AF) risk by using Mendelian randomization (MR) analysis.Methods: Single-nucleotide polymorphisms associated with vitamin D concentrations were used as instrumental variables.The outcome summary-level data for AF were obtained from the largest meta-analysis of genome-wide association studies consisting of 65,446 AF cases and 522,744 controls.Inverse-variance weighted (IVW) method was used as the primary analysis to estimate the causal effect.Robustness of the results was tested via sensitivity analyses by using multiple methods.Results:Genetically high 25-hydroxyvitamin D concentration was not associated with AF risk based on the results of the IVW analysis,with an odd ratio of 1.05 (95% confidence interval 0.84-1.31,P=0.667).Likewise,the results of the sensitivity analyses were consistent and supported our findings.MR-Egger regression indicated that directional pleiotropy did not bias the results (intercept=0.015,P=0.092).Conclusions: The MR analysis results do not support a causal association between vitamin D concentrations and AF risk.It is suggested that vitamin D supplementation may not reduce the risk of AF.关键词
