异银杏素对人肝癌Huh7细胞生物学行为的影响
Effect of isoginkgetin on biological behavior of human hepatoma Huh7cells

Abstract
Objective: To investigate the effect of isoginkgetin on the biological behavior of human hepatoma Huh7 cells. Methods:Human hepatoma Huh7 cells were exposed to differ-ent concentrations of isoginkgetin for a variety of durations. The cell viability was examined by CCK-8 assay. Scratch test and Transwell chamber assay were used to detect the cells'capacity of migration and invasion. The cell cycle and the apoptosis were detected by flow cytometry. Results:Isoginkgetin inhibited the proliferation of Huh7 cells in a dose-dependent manner(IC50=16. 68μmol/L,48 h;IC50=15. 42μmol/L,96 h). After exposure to isoginkgetin for 24 h,the rate of wound closure and invasion of the Huh7 cells decreased. The result of flow cytometry showed that the Huh7 cells treated with 12. 5μmol/L isoginkget-in were arrested in G0/G1 phases and the apoptosis rate in each group increased. Conclusion: Isoginkgetin could inhibit the proliferation,migration and invasion of Huh7 cells,change the cell cycle distribution and induce apoptosis.关键词
