miR-186-5p靶向HIF-1α对人骨肉瘤细胞存活、侵袭和迁移的影响
Effects of miR-186-5p targeting HIF-1α on survival, invasion and migration of human osteosarcoma cells

Abstract
Objective:To explore the effects of miR-186-5p and hypoxia inducible factor-1c (HIF-1α) on proliferation,apoptosis,invasion,and migration of human osteosarcoma cells.Methods:The expression levels of miR-186 in osteosarcoma tissues and cell lines were detected.Agomir-186-NC and Agomir-186 were transfected into U2OS cells,and the cells were randomly divided into three groups:Control group,Agomir-186-NC group and Agomir-186 group.The effects of overexpression of miR-186 on U2OS cellular proliferation,apoptosis,invasion,and migration were detected.The targeting relationship between miR-186-5p and HIF-1α was confirmed.Agomir-186 and shHIF-1α were transfected into U2OS cells alone or in combination.The cells were randomly divided into four groups:Control group,Agomir-186 group,shHIF-1α group,and Agomir-186+shHIF-1α group.The effects of miR-186 targeted HIF-1α on the proliferation,apoptosis,invasion,migration,and expression of relevant proteins in osteosarcoma cells were detected.Results:miR-186 was poorly expressed in human osteosarcoma tissues and cell lines(P<0.01).U2OS cellular proliferation multiple,number of invasive cells,and scratch closure rate were significantly reduced in the agomir186 group than those in the control group(P<0.01),while apoptosis rate was significantly increased(P<0.01).miR-186-5p was directly targeted to HIF-1α.The proliferation multiple,number of invasive cells,scratch closure rate,and the expression level of Ki67,N-cadherin and Vimentin protein were significantly reduced in the Agomir-186 and sh-HIF-1α co-transfection groups than those in the shHIF-1α group,while the apoptosis rate,the expression level ofCaspase-3 and E-cadherin were significantly increased(P<0.01).Conclusion:miR-186-5p can target HIF-1α,and inhibit the survival,invasion,and migration of human osteosarcoma cells.关键词
