夹竹桃活性内生真菌的分离筛选及菌株Aspergillus sp.次级代谢产物的研究
Isolation and screening of bioactive endophytic fungi from Nerium oleander and study on the secondary metabolites of Asper-gillus sp.

Abstract
Objective To isolate and screen the active endophytic fungi from Nerium oleander, and identify the structure and bioactivity of secondary metabolite of the fungi for obtaining bioactive compound. Methods The endophytic fungi were isolated through spread plate and streak plate methods. The target strain was selected under the guidance of bioassay through MTT method. Column chromatography on silica gel and semi-preparative HPLC were used to isolate and purify the metabolites from the fermentation of the selected fun-gas. The structures were identified by spectroscopy analysis and comparison with the data of literatures. Furthermore, the cytotoxicity of these compounds was evaluated. Results A total of 18 endophytic fungi were isolated from different parts of Nerium oleander Aspergillus sp. 10-4 metabolites showed cytotoxicity against human tumor H1975 cell lines. Two compounds were isolated from the ethyl acetate extract of Aspergillus sp. 10-4,and they were identified as 5-methoxysterigmatocystin and (+)-sydonic acid. Couclusion The 5-methoxysterigmatocystin is isolated from Nerium oleander-derived endophytic fungi for the first time. Endophytic fungi isolated from Nerium oleander have the potential of producing secondary metabolites with diverse structures.关键词
