摘要

Abstract

Objective: To comparatively study the effect and mechanism of Liuwei Dihuang Decoction with different preparations of Cornus officinalis on the expression of OPG/RANK/RANKL mRNA in femur of ovariectomized rats. Methods: 70 female SD rats were randomly divided into the blank group,the model group,the raw Cornus officinalis group,the wined Cornus officinalis group,the salt Cornus officinalis group, the steamed Cornus officinalis group and the control group (Alendronate Sodium ). The rat model of postmenopausal osteoporosis (PMOP) was established by ovariectomy. After 5 days of modeling, each group was given the corresponding intervention for 14 days. The body mass was measured once per week. 14 weeks later, radioimmunoassay was used to detect serum estradiol level,HE staining method was used to observe pathological changes of femoral head,and RT-qPCR method was used to detect mRNA expressions of OPG, RANK and RANKL in femur. Results: Compared to the model group,the Liuwei Dihuang Decoction groups with different preparations of Cornus officinalis could repair fractured trabecular bones of femoral head and reduce fat vacuoles; of which the effects of the wined group and the steamed group were better than the raw group and the salt group. Different Liuwei Dihuang Decoction groups could increase the serum level of estradiol (P<0.05),inhibit the body mass (P<0.05),increase the expression of OPG mRNA in femur (P<0.05),and reduce the expression of RANKL mRNA (P<0.05). Conclusion: Liuwei Dihuang Decoction with different preparations of Cornus officinalis can significantly improve PMOP, and its mechanism may be related to the increase of serum estrogen, regulate femur OPG/RANK/RANKL system,thereby interfere with bone metabolism and affect bone formation and bone resorption to improve PMOP. There were no significant differences in the effects of Liuwei Dihuang Decoction with different preparations of Cornus officinalis,but the wined Cornus officinalis and steamed Cornus officinalis had better effect. Clinically, appropriate preparations of Cornus officinalis should be chosen.
