姜黄素通过抑制HMGB1介导的自噬对棕榈酸诱导的卵巢颗粒细胞凋亡和类固醇生成能力的影响
Effect of curcumin on palmitic acid-induced apoptosis and steroid production of ovarian granulosa cells by inhibiting HMGB1-mediated autophagy

Abstract
Objective:To explore the effect of curcumin on the injury of ovarian granulosa cells induced by palmitic acid(PA) and its possible mechanism.Methods:Human ovarian granulosa cells were cultured in vitro.CCK-8 was used to de-tect cell viability.ELISA was used to detect estradiol (E2) con-tent in the cell supematant.RT-qPCR was used to detect the ex-pression of HMGB1,CYP19A and CYP11A mRNA.Flow cy-tometry was used to detect cell apoptosis.Western blotting was used to detect the expression of HMGB1,Beclinl,LC3B and P62 protein.Results:200 μmol/L or 400 μmol/L PA signifi-cantly decreased the cell viability,E2 content,and the expres-sion of CYP19A and CYP11A mRNA,while increased the apoptosis rate of cells (P<0.05).Compared with the control group,the cell viability,E2 content,and the expressions of CYP19A,CYP11A and P62 in the PA group were significantly reduced,while the cell apoptosis rate,the expressions of HMGB1 and Beclinl,and LC3Ⅱ/LC3Ⅰ ratio were significantly elevated (P<0.05).Compared with the PA group,the cell via-bility,E2 content,and the expressions of CYP19A,CYP11A and P62 in PA + curcumin (2.5 μmol/L or 5 μmol/L) group were increased,while the cell apoptosis rate,the expression of HMGB1 and Beclin1,and LC3Ⅱ/LC3Ⅰ ratio were evidently re-duced (P<0.05).Overexpression of HMGB1 reversed the pro-tective effects of curcumin on the PA induced injuries of ovari-an granulosa cells and up-regulated the expressions of autopha-gy-related proteins (P<0.05).Conclusion:Curcumin can at-tenuate PA-induced ovarian granulosa cell injury by inhibiting autophagy mediated by HMGB1.关键词
