High temperatures alter distribution of 5-methylcytosine on chromatin in Arabidopsis meiosis

Abstract
In many plant species,it has been evidenced that both generation and distribution of DSBs and COs are shaped by DNA methylation status of the chromatin. In Arabidopsis,elevation of environmental temperature over the tolerable range of DSB formation system influences CO distribution,and/or reduces CO formation by interfering with generation of DSBs and the assembly of synaptonemal complex. However, whether epigenetic modifications especially for DNA methylation of the chromosomes have an impact on response of MR to high temperatures remain unknown. To this end,a preliminary analysis of the distribution of 5-methylcytosine (5mC) was performed on prophase chromosomes in Arabidopsis plants incubated at high temperatures. Immunostaining using an anti-5mC antibody showed that under control temperature,5mC signals were dominantly located at heterochromatin regions where chromosomes showed a high staining florescence by 4',6-diamidino-2-phenylindole(DAPI). In contrast,under higher temperatures at both 32℃and 36-38℃, distribution of 5mC florescence is reshaped at different chromatin regions,suggesting that DNA methylation landscape be altered under the increased temperatures. This study therefore hinted that elevation of environmental temperatures may affect DSB and/or CO patterning by modifying DNA methylation status.关键词
