Effect of down-regulation of ADAM17 on natural killer cell killing in lung squamous cell carcinoma

Abstract
Objective:To investigate the effect of ADAM17 down-regulation on killing effect of natural killer cells(NK)in lung squamous cell carcinoma.Methods:Human NCI-H520 cell lines were cultured in vitro and they were divided into normal control group(non-transfected),negative control group(transfected nonsense siRNA)and ADAM17 down-regulated group(transfected ADAM17 siRNA).Real-time quantitative fluorescent polymerase chain reaction(qRT-PCR)was used to detect ADAM17 mRNA expression in NCI-H520 cells of each group,and Western blotting was used to detect ADAM17 protein expression in NCI-H520 cells of each group.NK cells were prepared and added into NCI-H520 cells of each group to detect the killing activity of NK cells.Western blotting detected the expression of γ interferon(IFN-γ),granulozyme B(GraB)and perforin(PFP)proteins in the cells of each group.Results:Compared with normal control group,the ADAM17 mRNA and protein relative expression levels of negative control group were with no significant differences;and ADAM17 down-regulated group significantly decreased the mRNA and protein relative expression levels of ADAM17(P＜0.01);ompared with normal control group,the survival rate of NCI-H520 had no significant differences;the survival rate of NCI-H520 in ADAM17 down-regulated group were significantlly decrased(P＜0.01);compared with normal control group,the expression of IFN-γ、GraB、PF in negative control group had no significant differences,and the relative expression levels of IFN-γ,GraB and PFP protein were significantly increased(P＜0.01).Conclusion:ADAM17 down-regulation in NCI-H520 lung squamous cell carcinoma can enhance the killing effect of NK cells.关键词
