鸡矢藤醇提物联合抗生素对MRSA的体外抑菌效果研究
Study on the in vitro antibacterial effect of ethanol extract from Paederia scandens combined with antibiotics on MRSA

Abstract
Objective To explore the antibacterial effect of ethanol extract of Paederia scandens(Lour)Merrill(Rubiaceae)and its combination with five commonly used antibiotics including polymyxin B,levofloxacin,gentamicin,ampicillin and vancomycin on Methicillin-resistant Staphylococcus aureus(MRSA).Methods Oxford cup method was used to determine the antibacterial effect of ethanol extract of Paederia scandens alone on MRSA clinical strains,MRSA standard strain ATCC43300,and Staphylococcus(S.)aureus strain ATCC25923.The micro broth dilution method was used to determine the minimum inhibitory concentration(MIC)and minimum bactericidal concentration(MBC)of Paederia scandens ethanol extract and the five antibiotics on clinical and standard strains of MRSA,and S.aureus ATCC25923.Micro-dilution chessboard method was used to determine the synergistic antibacterial effect of Paederia scandens ethanol extract and the five antibiotics.Results The MIC of Paederia scandens ethanol extract against both ATCC25923 and ATCC43300 were 125.0 mg/mL,the MBC both were 500.0 mg/mL.The MIC of Paederia scandens ethanol extract against clinical MRSA strains ranged from 62.5-250.0 mg/mL,the MBC ranged from 250.0-500.0 mg/mL.In addition,when combined with gentamicin,Paederia scandens ethanol extract had a synergistic effect against ATCC43300(FIC=0.375).Additive effects were observed when combined with levofloxacin,ampicillin,and vancomycin against MRSA ATCC43300(FIC=0.625,FIC=0.531,and FIC=0.531,respectively).However,no effect was observed when combined with polymyxin B against ATCC43300.When tested against clinical strains of MRSA,Paederia scandens ethanol extract showed an additive effect when combined with polymyxin B,ampicillin,levofloxacin,and gentamicin(FIC =0.625,FIC=1.0,FIC=0.531,and FIC=0.75,respectively),and no effect was observed when combined with vancomycin.Furthermore,the extract showed an additive effect when combined with levofloxacin,gentamicin,and vancomycin against ATCC25923(FIC=1.0,FIC=0.625,and FIC=0.516,respectively).Conclusion Paederia scandens ethanol extract has a significant inhibitory effect on both clinical and standard strains of MRSA and S.aureus.The combination of the ethanol extract with commonly used antibiotics showed effectiveness in treating these bacteria,suggesting that the natural antibacterial components of Paederia scandens ethanol extract may offer new possibilities in addressing the issue of antibiotic resistance.关键词
