Effects of Transgelin-2 silencing on malignant biological behavior of e-sophageal squamous cell carcinoma cells infected by Porphyromonas gin-givalis

Abstract
Aim:To investigate the effects of silencing Transgelin-2 on the malignant biological behavior of esophageal squamous cell carcinoma(ESCC)cells infected by Porphyromonas gingivalis(PG)and its possible mechanism.Methods:NE6-C or KYSE140 cells were allocated into 4 groups and given the following treatments:transfected with siControl,PG in-fection(multiplicity of infection 10),siTAGLN-2 transfection,and siTAGLN-2 transfection +PG infection,respectively.Cell proliferation was detected by CCK-8,cell cycle was detected by BrdU mark,migration and invasion were detected by Tran-swell chamber assay,and the distribution of Transgelin-2,F-actin,E-cadherin and Vimentin proteins was detected by immu-nofluorescence assay,the protein expressions of Transgelin-2,E-cadherin and Vimentin were detected by Western blot.Re-sults:PG infection alone increased the proliferation rate of NE6-C and KYSE140 cells,increased the proportion of S-phase cells,the number of invasive cells and the number of migratory cells(P<0.05),and up-regulated Transgelin-2 expression(P<0.05),decreased F-actin;down-regulated E-cadherin expression and up-regulated Vimentin expression(P<0.05).siTAGLN-2 transfection significantly antagonized the above effects of PG infection(P<0.05).Conclusion:Transgelin-2 may inhibit the malignant progression of ESCC cells induced by PG infection through altering the cytoskeleton.关键词
