鼠源M2型巨噬细胞对KYSE30细胞克隆、侵袭、迁移及EMT相关蛋白表达的影响
Effects of murine M2 type macrophages on clone,invasion,migration and expressions of EMT related proteins of KYSE30 cells

Abstract
Aim:To explore the effects of murine M2 type macrophages on the colony formation,invasion,migration and the expressions of epithelial mesenchymal transition(EMT)-related proteins of esophageal squamous carcinoma KYSE30 cells.Methods:Murine macrophages RAW264.7 were divided into induction group and blank control group.The induction group was cultured in DMEM medium containing 0.02 mg/L IL-4 and IL-13.The blank control group was cultured in DMEM medium.The macrophages in the 2 groups were cultured for 48 hours.The percentage of CD206+F4/80+ macropha-ges was detected by flow cytometry.The expressions of CD206,Mgl-2,Clec10a mRNA in the cells were detected by qRT-PCR.The protein expression of CD206 in the cells was detected by Western blot.KYSE30 were divided into control group(only cultured)and co-culture group.The co-culture group cells were cultured with Murine M2 type macrophages.The clone formation ability of cells in the 2 groups were detected by plate colony formation assay.The protein expressions of E-cadher-in,N-cadherin,Vimentin and MMP-9 in the 2 groups were detected by Western blot.The invasion and migration ability in the 2 group were detected by Transwell chamber and wound healing assay.Results:Compared with blank control group,the percentage of CD206+F4/80+,the mRNA expressions of CD206,Mgl-2,Clec10a and the protein expression of CD206 in the induction group increased(P<0.05).Compared with control group,the ability of colony formation in the co-culture group enhanced(P<0.001),the expressions of E-cadherin,N-cadherin,Vimentin and MMP-9 increased(P<0.05)and the ability of invasion and migration enhanced(P<0.001).Conclusion:Murine M2 type macrophages after induction can enhance the ability of colony formation,invasion,migration and the expressions of EMT-related proteins of KYSE30.关键词
