Abstract

In 2023,the composite enzyme substrate method is included in the Standard Examination Methods for Drinking Water——Part 12:Microbial Indeices(GB/T 5750.12-2023)as a new method for detecting the total number of bacterial colonies in water.In order to facilitate more scientific and standard use of complex enzyme substrate method to detect the total number of colonies in drinking water,this paper adopted complex enzyme substrate method to detect 80 groups of actual samples of drinking water and source water,and analyzed the detection results of two different culture times of 24 h and 48 h.The test results showed that there was a significant difference between the 24 h culture results of SimPlate method and 48 h culture results.The paired t-test analysis showed that the 24 h culture results of enzyme substrate method and 48 h culture results P=0.000(<0.05),which had a significant statistical difference,It was found that some pathogenic bacteria could not be detected by SimPlate culture for 24 h.Therefore,when enzyme substrate method was used to detect the total number of colonies in drinking water and source water,the culture time should be at least 48 hours to meet the detection requirements of the laboratory on the total number of colonies in drinking water and source water.
