沙库巴曲缬沙坦对血管平滑肌细胞增殖的影响
EFFECT OF ENTRESTO ON THE PROLIFERATION OF VASCULAR SMOOTH MUSCLE CELLS

Abstract
Objective To investigate the effect of Entresto(sacubitril/valsartan)on the proliferation of vascular smooth muscle cells(VSMCs)stimulated by angiotensin Ⅱ(Ang Ⅱ).Methods Human aortic VSMCs were cultured in vitro and ran-domly divided into Ang Ⅱ group(treated with Ang Ⅱ),Ang Ⅱ+Entresto group(treated with Entresto for 1 h,followed by AngⅡ treatment after medium change),and control group(without drug treatment).All groups were cultured for 24 h.CCK8 assay was used to measure cell proliferation;scratch assay was used to measure cell migration activity;quantitative real-time PCR and Western blot were used to measure the expression levels of proliferating cell nuclear antigen(PCNA)and matrix metalloproteinase-9(MMP-9).Results Compared with the control group,the Ang Ⅱ group had significant increases in cell proliferation rate,mi-gration activity,and the expression levels of PCNA and MMP-9 in VSMCs(F=16.54-177.20,P＜0.05).Compared with the Ang Ⅱ group,the Ang Ⅱ+Entresto group had significant reductions in cell proliferation rate,migration activity,and the expres-sion levels of PCNA and MMP-9 in VSMCs(P＜0.05).Conclusion Entresto can inhibit the proliferation and migration of VSMCs induced by Ang Ⅱ.关键词
