胃癌患者癌组织和血清外泌体中LncRNA CCAT1、miR-140-3p表达及其临床意义
Expression and clinical significance of LncRNA CCAT1 and miR-140-3p in cancer tissues and serum exosomes of patients with gastric cancer

Abstract
Objective To observe the expression and clinical significance of long non-coding ribonucleic acid colon cancer associated transcript1(LncRNA CCAT1)and microRNA-140-3p(miR-140-3p)in the cancer tissues and serum exosomes of patients with gastric cancer.Methods Ninety-three gastric cancer patients who underwent radical gastrecto-my were selected as the gastric cancer group,while 47 patients with benign gastric lesions were selected as the benign le-sion group,and 47 patients with healthy physical examination were selected as the healthy control group during the same period.The expression of LncRNA CCAT1 and miR-140-3p in the gastric cancer tissues,adjacent tissues and serum exo-somes of all subjects were detected by reverse transcription quantitative real-time polymerase chain reaction(RT-qPCR).The gastric cancer patients were followed up for 5 years,and we recorded their survival time and the 5-year survival rate.Pearson correlation analysis was used to analyze the correlation between the LncRNA CCAT1 and miR-140-3p in the gas-tric cancer tissue and serum exosomes of gastric cancer patients.Based on the average expression of LncRNA CCAT1 and miR-140-3p in the cancer tissue of gastric cancer patients,patients were divided into the high and low LncRNA CCAT1 and miR-140-3p individuals.K-M survival curve was used to analyze the 5-year survival rates of high and low LncRNA CCAT1 and miR-140-3p patients.The diagnostic value of serum exosomes LncRNA CCAT1 and miR-140-3p in the gastric cancer patients were analyzed by receiver operator characteristic(ROC)curve.Results The expression of LncRNA CCAT1 in the gastric cancer tissues of patients was higher than that in the adjacent tissues,and the expression of miR-140-3p was lower than that of the adjacent tissues(both P<0.05).The expression of LncRNA CCAT1 in the serum exosome were as follows:the healthy control group<benign lesion group<gastric cancer group,and the miR-140-3p expression was in the following order:the healthy control group>benign lesion group>gastric cancer group(all P<0.05).Pearson correla-tion analysis showed that there was a negative correlation between the expression of LncRNA CCAT1 and miR-140-3p in the cancer tissues and serum exosomes of gastric cancer patients(r=-0.726,-0.639,respectively;all P<0.05).There were statistically significant differences in the expression levels of LncRNA CCAT1 and miR-140-3p in the gastric cancer tissues and serum exosomes of gastric cancer patients with different levels of tumor differentiation,depth of invasion,clini-cal stage,and lymph node metastasis(all P<0.05).The 5-year overall survival rate of gastric cancer patients with high ex-pression of LncRNA CCAT1 in the cancer tissues and serum exosomes was lower than that of patients with low expression of LncRNA CCAT1,and the 5-year overall survival rate of gastric cancer patients with high expression of miR-140-3p was higher than that of patients with low expression of miR-140-3p(both P<0.01).The ROC analysis results showed that the area under the curve(AUC)of serum exosomes LncRNA CCAT1 and miR-140-3p in predicting gastric carcinoma was 0.712 and 0.766,respectively;the AUC of them combined in diagnosis of gastric cancer was 0.845.Conclusion The relative expression of LncRNA CCAT1 in the gastric cancer tissues and serum exosomes increased,while the relative ex-pression level of miR-140-3p decreased;their expression levels were related to the degree of tumor differentiation,depth of infiltration,clinical staging,lymph node metastasis,and prognosis of patients,and the combination of serum exosomes Ln-cRNA CCAT1 and miR-140-3p had high diagnostic value for gastric cancer.关键词
