急性胰腺炎小鼠胰腺和远隔脏器组织TRAF6、XIAP表达变化及其与细胞凋亡的关系
Expression changes of TRAF6 and XIAP in pancreas and distal organs of mice with acute pancreatitis and their relationship with apoptosis

Abstract
Objective To observe the expression changes of tumor necrosis factor receptor-associated factor 6(TRAF6)and X-linked apoptosis inhibitory protein(XIAP)in the pancreas and distal organs of mice with acute pancreati-tis and to analyze their relationship with apoptosis in multiple organs.Methods Thirty male C57BL/10SnJ mice were randomly divided into the control,acute edematous pancreatitis(AEP),and acute necrotizing pancreatitis(ANP)groups,with 10 mice in each group.In the AEP group,we established the AEP models;in the ANP group,we established the ANP models;in the control group,mice were injected with an equal volume of phosphate-buffered saline.TRAF6 and XIAP mRNA in pancreas,intestine,liver,lung and kidney tissues of each group were detected by RT-qPCR,and cleaved Caspase-3,TRAF6 and XIAP proteins were detected by Western blotting.The apoptosis rates of pancreas,small intes-tine,liver,lung and kidney tissues were measured by TUNEL method.Results Statistically significant differences were found in the TRAF6 and XIAP protein expression in the pancreas and several distal organs between the ANP and AEP groups and the control group(all P<0.05),XIAP mRNA expression in the pancreatic tissues of the AEP group was lower than that in the ANP group(P<0.05),TRAF6 protein expression in the pancreatic,intestinal,hepatic,lung,and renal tissues of the AEP group was higher than that of the ANP group,and XIAP protein expression was lower than that in the ANP group(all P<0.05).The apoptosis rate and cleaved Caspase-3 expression in the pancreatic,intestinal,liver,lung and kidney tissues in the AEP group were higher than those in the ANP group and the control group(all P<0.05).The ex-pression of cleaved Caspase-3 protein in the pancreas and several distal organs of pancreatitis mice was positively correlat-ed with TRAF6 protein expression and was negatively correlated with XIAP protein expression(all P<0.05).Conclusions The expression of TRAF6 and XIAP in the pancreas and distal organ tissues of mice with acute pancreatitis is abnormal and correlated with apoptosis indexes;the expression trends of RAF6 and XIAP in AEP and ANP are different,and the regula-tory effects of RAF6 and XIAP on apoptosis may be related to the severity of acute pancreatitis.关键词
