Bioactive polyphosphate biomaterials for bone regeneration:Fundamental,progress and challenge

Abstract
Functional orthopedic biomaterials are emerging in accordance with a better understanding of bone regeneration and progress of tissue engineering scaffolds.Inorganic polyphosphate(polyP)is a physiologically occurring biopolymer with high-energy phosphoric anhydride bonds that exists in osteoblasts and platelets,whose degradation products such as phosphate ions and energy can participate in bone regeneration and metabolism.Inspired by such events,polyphosphate biomaterials have been explored as novel bone substitutes in both amorphous and crystalline forms,along with impressive outcomes both in vitro and in vivo.This review summarizes the characteristics,physiological effects,cellular mechanisms,and material science progress of such unique biomaterial,providing basis of the application of high-energy polyphosphate in bone regeneration.In spite of its fundamentals and advances,challenges and controversies of polyphosphate are also discussed as an effort to promote its bench to bedside clinical translation in the future.关键词
