Abstract

Objective To investigate the effect of Anillin actin-binding protein(ANLN)gene knockout on the proliferation,migration and behavior of lung adenocarcinoma(LUAD)cells.Methods ANLN-siRNA plasmid was constructed and transfected into A549 lung adenocarcinoma cells by electroporation.After mono-clonal selection,identification and expansion culture,the experimental group(the ANLN-A549 group)was ob-tained.The control group(the Ctrl-A549 group)was obtained by transfecting A549 cells with empty vector plasmid.Transwell cell migration assay and cell scratch test were used to detect the migration of lung cancer cells in the two groups,and CCK-8 cell proliferation assay and cell colony formation assay were used to detect the clonal proliferation ability of lung cancer cells.Results Transwell cell migration and cell scratch experi-ments showed that after ANLN gene knockout,the migration rate of lung cancer cells in the ANLN-A549 group was significantly lower than that in the Ctrl-A549 group,and the difference was statistically significant(P<0.05).The results of CCK-8 cell proliferation assay showed that the proliferation of ANLN-A549 group was significantly slower than that of the Ctrl-A549 group(P<0.05).Cell colony formation assay showed that the number of cell communities in the ANLN-A549 group was significantly lower than that in the Ctrl-A549 group(P<0.05).Conclusion The knockout of ANLN gene significantly inhibits the proliferation and migra-tion of lung adenocarcinoma cells.
