Determination of Three Components in Xiaoer Feire Kechuan Granules by HPLC

Abstract
Objective:To develop an HPLC method for simultaneous quantitation of chlorogenic acid,forsythoside A and baicalin in Xiaoer Feire Kechuan Granules by HPLC.Methods:The chromatographic column was Agilent Eclipse Plus C18(250 mm×4.6 mm,5 pm)with gradient elution by methanol-0.2％formic acid solution.The detection wavelengths were 327 nm.The column temperature was 35 C and the mobile phase flow rate was 1.0 mL/min.The sample injection volume was 10 pL.Results:Chlorogenic acid,forsythoside A and baicalin showed good linear relationships within the range of 2.69～134.30 pg/mL(r=0.9998),11.81～590.60 pg/mL(r=0.9995)and 21.44～1072.25 pg/mL(r=0.9998),respectively,and the average recoveries were 99.37％with RSD of 1.20％(n=6),98.14％with RSD of 1.57％(n=6)and 99.14％with RSD of 1.66％(n=6).Conclusion:The method is highly specific,and can be used to detect the content of main decoction pieces such as Lonicerae Japonicae Flos,Forsythia Fructus and Scutellariae Radix,and can be used for the overall quality control of Xiaoer Feire Kechuan Granules.关键词
