摘要
Abstract
To analyze the application of 2,3,5-triphenyltetrazolium chloride(TTC)as a bacterial culture indicator in Mycoplasma culture.Mycoplasma bovis,Mycoplasma mycoides subsp.capri,Mycoplasma capricolum subsp.capripneumoniae,Mycoplasma ovipneumoniae,Mycoplasma gallisepticum and Mycoplasma pneumoniawere cultured in modified Thiaucourt's liquid culture medium(MTB)and modified Thiaucourt's solid culture medium(MTA)with TTC,and MTB and MTA added the phenol red indicator as culture control.The growth of Mycoplasma was observed in both TTC medium and phenol red medium,then the optimal concen-tration of TTC was screened.The results showed that TTC indicator exhibited more sensitive than con-ventional phenol red in the culture medium of Mycoplasma.In MTA medium with TTC as indicator,Mycoplas-mal colonies were more valuable for observing and counting colony-forming units(CFU).The optimal con-centration of TTC in Mycoplasma medium was 0.2 mg/mL.This study will provide basic on the optimization of Mycoplasma culture and vaccine production process.关键词
