Effect of Electroacupuncture at Shenmen Point on Behavioral Parameters and FK506 Binding Protein 5 Expression in the Prefrontal Cortex in Mice with Posttraumatic Stress Disorder

Abstract
Objective To investigate the effect of electroacupuncture at Shenmen point on anxiety-like behavior and FK506 binding protein 5(FKPB5)expression in the prefrontal cortex in mice with posttraumatic stress disorder(PTSD).Methods C57 mice were randomly divided into normal group,model group,and electroacupuncture group,with 6 mice in each group.The mice in the model group and the electroacupuncture group were given single prolonged stress and electrical stimulation to establish a mouse model of PTSD,and then the mice in the electroacupuncture group were given electroacupuncture at Shenmen point for 15 mi-nutes each time,once a day for 14 consecutive days.The open field test and the elevated plus maze test were used to measure the behavioral parameters of mice.Quantitative real-time PCR and Western blot were used to measure the mRNA and protein ex-pression levels of FKPB5 in the prefrontal cortex.Results The open field test showed that electroacupuncture significantly im-proved the retention time in the central area and the number of squares crossed(P<0.05),and the elevated plus maze test showed that electroacupuncture significantly improved the open arm time,number of open arm entries,percentage of open arm time,and percentage of open arm entries(P<0.05).Compared with the normal group,the model group had significant increa-ses in the protein and mRNA expression levels of FKBP5 in the prefrontal cortex(P<0.05),and compared with the model group,the electroacupuncture group had significant reductions in the protein and mRNA expression levels of FKBP5 in the pre-frontal cortex(P<0.05).Conclusion Electroacupuncture can significantly improve the anxiety-like behavior of mice with PTSD,possibly by increasing the expression level of FKBP5 in the prefrontal cortex.关键词
