番茄红素对Fscn2基因敲除小鼠听力的保护作用
Effects of lycopene on the hearing impairment in Fscn2-/-mice

Abstract
Objective To investigate the potential protective effect of lycopene on hearing impairment in Fscn2-/-mice.Meth-ods Fscn2-/-mice were genotyped by PCR.Fscn2-/-mice at age of 1 week were randomly divided into 2 groups and treated as follows:the experimental group(n =10)received lycopene(diluted with corn oil)of20 μg/g by intraperitoneal injection,once every other day for35 days;the control group(n =10)received similar amount of corn oil(1.0 μL/g)at the corresponding time points.Auditory brainstem response(ABR)thresholds were measured in mice 48 h after the last injection.Moreover,the morphological structure and number of outer hair cells stained by phalloidin were observed in the mouse cochleae.Additionally,the expression levels of Bax,Bcl-2 and Cleaved caspase12 in the inner ears were detected by Western blot.Results ABR thresholds of the mice treated with lycopene(20 μg/g)were decreased at all the stimulus frequencies(P<0.05 for 8 kHz,16 kHz and click).Staining of hair cell stereocilia revealed that loss of outer hair cells in mice treated with lycopene was significant-ly less than that of the control mice at the basal turns(P<0.05).Western blot showed that,after intraperitoneal injection of ly-copene,the levels of apoptosis-related molecules(Bax and Cleaved caspase 12)were decreased,whereas the level of anti-ap-optotic protein(Bcl-2)was increased.Conclusion Administration of lycopene protects against hearing impairment in Fscn2-/-mice.Anti-apoptosis is likely involved in the oto-protective effects of lycopene.关键词
