Abstract
Objective To investigate the hepatoprotective activities of Dendrobium huoshanense C.Z.Tang et S.J.Cheng(DH)in mouse model of high-fat-sucrose diet(HFD)-induced non-alcoholic fatty liver disease(NAFLD).Methods NAFLD model was induced in C57BL/6 mice by feeding with high-fat diet for 12 weeks.Mice were randomly divided into five groups:the control group,the NAFLD model group,the 0.1 mg/kg DH group,the 1 mg/kg DH group and the 10 mg/kg DH group.Levels of alanine aminotransferase(ALT),total triglycerides(TG),total cholesterol(TC),low-density lipoprotein cholesterol(LDL-C)and aspartate aminotransferase(AST)in serum were analyzed by ELISA.Liver ultrastructure was observed via optical mi-croscopy with HE staining method.Oil red O-staining method was used to detect the lipid content,and Sirius red staining method was used to observe liver fibrosis.CD68 expression in liver tissues and serum levels of inflammatory factor(IL-1β,IL-6 and TNF-ɑ)were analyzed to define inflammation status.Results Compared with those in the control group,levels of AST,ALT,TG,TC,and LDL-C were increased,and severe liver damage,lipid accumulation and liver fibrosis were observed in the NAFLD model group.In addition,the mice in the NAFLD model group also showed severe inflammatory response,mainly manifested as significantly increased levels of IL-1β,IL-6 and TNF-ɑ in serum and significantly increased expression of CD68 in liver tissues.Compared with the NAFLD model group,DH treatment could significantly alleviate these liver injury manifestations in NAFLD mice.Conclusion Dendrobium huoshanense can effectively reduce high-fat diet-induced non-alcoholic fatty liver injury in mice,and this hepatoprotective effect may be related to its anti-inflammatory effect and lipid-lowering effect.关键词
