群体感应淬灭酶AiiO的克隆表达及其发酵工艺优化

摘要
Abstract
The quorum sensing quenching enzyme AiiO has the activity of degrading signal molecule N-acyl homoserine lactone(AHL),inhibiting the pathogenicity of pathogens.To increase the expression level of AiiO,a recombinant strain Escherichia coli BL21(DE3)-pET-28a(+)-aiiO was constructed.The induction temperature,nitrogen source content,induction timing,and lactose addition during its fermentation production of AiiO were optimized.The results showed that under the optimized conditions of inducing temperature of 25 ℃,peptone of 20 g/L and yeast powder of 10 g/L,adding lactose at the beginning of fermentation,and lactose addition of 5 g/L,the expression level of intracellular soluble AiiO reached 374.26 mg/L.关键词
