Abstract

The effects of ultrasound power,extraction time,ethanol volume fraction,and solid-liquid ratio on the extraction process of resveratrol from Polygonum cuspidatum were investigated using single factor and response surface experiments.A combination of macroporous adsorption resin and reverse phase column chromatography was used to separate and identify resveratrol by HPLC.The intracellular and extracellular antioxidant studies were conducted,as well as protective effects on fibroblasts under UV irradiation.The optimal extraction conditions for resveratrol were ultrasound power of 496 W,extraction time of 33 min,ethanol volume fraction of 64%,and solid-liquid ratio of 1∶17.Under the optimal conditions,the extraction rate was 4.80 mg/g and the purity of the obtained resveratrol reached 99.90%.When the contents of resveratrol were 2.5 and 0.5 mg/mL,DPPH and hydroxyl radical scavenging rates reached the highest of 94.92%and 94.65%,and the IC50 were 0.926 and 0.165 mg/mL,respectively.Resveratrol can significantly reduce SOD,CAT,GSH and MDA in skin fibroblasts exposed to ultraviolet radiation.
