益气活血方对急性心肌梗死大鼠交感神经重构的影响
Effect of Yiqi Huoxue Formula(益气活血方)on Sympathetic Nerve Remodeling in Rats with Acute Myocardial Infarction

Abstract
Objective:To observe the effect of Yiqi Huoxue formula on sympathetic nerve remodeling in rats with acute myocardial infarction during acute cardiac injury and explore its mechanism.Methods:The acute myocardial infarction model of SD rats was established by ligating the anterior descending branch of the left coronary artery in rats.Before modeling,20 rats were randomly selected as the sham operation group without ligation.The successful rats were randomly divided into model group,Yiqi Huoxue formula group and metoprolol group,with 20 rats in each group.Rats in Yiqi Huoxue formula group and metoprolol group were respectively intragastrically administered with corresponding drugs,and rats in model group were intragastrically administered with distilled water after 24 hours of modeling.Cardiac function,susceptibility to ventricular fibrillation,patho-logical morphology of myocardial tissue,distribution of tyrosine hydroxylase(TH)-positive sympathetic nerve fibers,plasma neuropeptide Y(NPY)content and protein expression of TH,growth associated protein 43(GAP43),nerve growth factor(NGF)and Sema-3A were detected after 3 and 7 days of administration.Results:On the 3 rd and 7th day after administration,the model group showed lower LVEF and LVFS than sham operation group(P<0.01),while higher LVIDd and LVIDs than sham operation group(P<0.01).On the 3rd day after administration,Yiqi Huoxue formula group showed lower LVIDs than model group(P<0.05).On the 7th day after administration,the Yiqi Huoxue formula group and metoprolol group showed higher LVEF and LVFS than model group(P<0.05 or P<0.01),while lower LVIDs than model group(P<0.05).The LVIDs of metoprolol group after 7 days of administration was lower than those after 3 days of administration(P<0.05).On the 3rd and 7th day after adminis-tration,the model group showed lower threshold of ventricular fibrillation induced by electrical stimulation,than sham operation group(P<0.01 or P<0.05).On the 7th day after administration,the metoprolol group showed higher threshold of ventricular fibrillation induced by electrical stimulation,than model group(P<0.01).The threshold of ventricular fibrillation induced by electrical stimulation in metoprolol group after 7 days of administration was lower than those after 3 days of administration(P<0.05).After 3 and 7 days of administration,the myocardial cells were broken and arranged in disorder in model group,with pyknosis of nuclei and infiltration of a large number of inflammatory cells.After 3 days of administration,the myocardial cell structure was disordered and partially necrotic in the Yiqi Huoxue formula group and metoprolol group.After 7 days of administration,the Yiqi Huoxue formula group improved significantly compared with the model group,and the arrangement of myocardial cells in metoprolol group was significantly changed.After 3 and 7 days of administration,the density of TH positive nerve fibers increased significantly in the model group and the distribution was disordered.After 3 days of administration,the Yiqi Huoxue formula group and metoprolol group showed lower density of TH positive nerve fibers than model group.After 7 days of administration,the nerve morphology and distribution tended to normalize in the Yiqi Huoxue formula group and metoprolol group,and the Yiqi Huoxue formula group showed less positive nerve fibers than metoprolol group.After 3 and 7 days of administration,the model group showed higher plasma NPY content than sham operation group(P<0.01).On the 7th day after administration,the Yiqi Huoxue formula group and metoprolol group showed lower plasma NPY content than model group(P<0.01).The plasma NPY content in Yiqi Huoxue formula group and metoprolol group after 7 days of administration was lower than those after 3 days of administration(P<0.01).On the 3rd and 7th day after administration,the model group showed higher relative expressions of NGF,GAP43 and TH proteins than sham operation group(P<0.01),while lower relative expression of Sema-3A protein than sham operation group(P<0.01).On the 3rd and 7th day after adminis-tration,the Yiqi Huoxue formula group and metoprolol group showed lower relative expression levels of NGF,GAP43 and TH proteins than model group(P<0.05),while higher relative expression levels of Sema-3A protein than model group(P<0.05).On the 3rd day after administration,the Yiqi Huoxue formula group showed higher relative expression of NGF protein than metoprolol group(P<0.05).On the 7th day after administration,Yiqi Huoxue formula group showed higher the relative expression of TH protein than the metoprolol group(P<0.05).Conclusion:Yiqi Huoxue formula may increase the threshold of ventricular fibrillation induced by electrical stimu-lation in rats with acute myocardial infarction,reduce the protein expression levels of GAP43,NGF and TH,increase the protein expression level of Sema-3A,and inhibit sympathetic nerve remodeling in the early stage of myocardial infarction.关键词
