ANXA6基因单核苷酸多态性及其表达水平与慢性牙周炎的关联性研究
Relationship between ANXA6 gene single nucleotide polymorphisms and its expression and chronic periodonti-tis

Abstract
Objective:To investigate the relationship between AnnexinA6(ANXA6)gene single nucleotide polymor-phism(SNP)and its expression level and chronic periodontitis.Methods:Totally 142 patients with chronic periodontitis who were treated in Department of Stomatology,Second Affiliated Hospital of Xinjiang Medical University from January 2022 to December 2022 were selected as the study objects,and 150 healthy subjects who underwent physical examination in our hos-pital during the same period were selected as the healthy control group.Two SNP loci rs11960458 and rs4958897 of ANXA6 gene were identified by capillary electrophoresis and fragment analysis(SNaPshot).The relative mRNA expression of ANXA6 gene was determined by qRT-PCR.Results:No significant differences were found in gender,age,BMI,smoking history and drinking history between chronic periodontitis patients and healthy control group(P>0.05).For rs11960458,there were sig-nificant differences in CC,CT and TT genotypes between the chronic periodontitis group and the healthy control group(P= 0.002).The comparison of additive model and dominant model between the two groups was statistically significant(P<0.05).For rs4958897,there were no significant differences in CC,CT and TT genotype distribution and genetic model distri-bution between chronic periodontitis group and healthy control group(P>0.05).There was significant difference in allele fre-quencies of C and T genes at rs11960458(P=0.005),but no significant difference in allele frequencies of C and T genes at rs4958897(P=0.089)between the two groups.The relative mRNA expression levels of ANXA6 gene in chronic periodontitis patients were significantly higher than those in healthy controls(P<0.05).The area under the curve(AUC)to distinguish chronic periodontitis patients from healthy controls using ANXA6 gene mRNA relative expression level was 0.876(95%CI 0.835～0.916,P<0.001).The sensitivity and specificity were 72.5%and 89.3%respectively.The relative mRNA expres-sion levels of ANXA6 gene in patients with CT and TT genotype at rs11960458 were significantly higher than those in patients with CC genotype(P<0.05),but there was no significant difference in the relative mRNA expression levels of ANXA6 gene in patients with CT and TT genotype(P>0.05).Conclusion:The variation of rs11960458 in ANXA6 gene and the level of ANXA6 gene expression are related to the onset of chronic periodontitis.关键词
