Abstract
Aim To establish a macrophage-like phenotypic transformation model of human vascular smooth muscle cells(VSMC)induced by water-soluble cholesterol,and to explore the role of melatonin in this process.Meth-ods VSMC is divided into a control group,a cholesterol group(water-soluble cholesterol),and a melatonin group(water-soluble cholesterol+melatonin).Establishment of a macrophage like phenotype transformation model in VSMC induced by water-soluble cholesterol.The expression of macrophage-like phenotypic related genes in VSMC was detected by quantitative PCR.Oil red O staining experiment was conducted to observe the effect of melatonin on the lipid accumulation of VSMC in-duced by water-soluble cholesterol.Edu staining was used to observe the effect of melatonin on the proliferation of VSMC induced by water-soluble cholesterol.The effect of melatonin on VSMC migration induced by water soluble cholesterol was evaluated by cell scratch assay.Results The mRNA expression of smooth musclecontraction phenotype related mark-ers,α-actin and α-tropomyosin,were significantly decreased after 10 mg/L water-soluble cholesterol treatment,while the mRNA expression macrophage related markers,CD68 and MAC2,were significantly increased after treated by 10 mg/L wa-ter-soluble cholesterol,suggesting the transformation of VSMC to a macrophage-like phenotype.In addition,0.5 μmol/L melatonin inhibited lipid accumulation and proliferation of VSMC induced by water-soluble cholesterol,but had no effect on VSMC migration.Conclusion Melatonin inhibits the macrophage-like phenotype transformation of VSMC.关键词
