花姜酮通过抑制NLRP3炎症小体改善SAP大鼠肠道黏膜屏障功能
Zingerone ameliorates intestinal mucosal barrier function in SAP rats by inhibiting NLRP3 inflammasome

Abstract
Aim To explore the effect of zingerone on the intestinal mucosal barrier function of rats with severe acute pancreatitis(SAP).Methods 42 SD rats were randomly divided into the control group(SD rats treated with distilled water),the model group(SAP rats treated with distilled water),and the zingerone group(SAP rats treated with zingerone).The histopathological changes of the small intestine were detected by HE staining;Interleukin(IL)-1β and tumor necrosis factor-α(TNF-α)in serum and small intestine tissue were detected by ELISA.Nitric oxide(NO)level was measured by Griess method.Endotoxemia(ET)content was measured by azo matrix color method.Latex enhance-ment immunoturbidimetric method was used to determine the content of myeloperoxidase(MPO).EB staining was used to determine intestinal vascular permeability.Immunofluorescence method was used to detect the number of nucleotide-bind-ing oligomerization domain-like receptor protein 3(NLRP3)and apoptosis-associated speck-like protein containing CARD(ASC)positive cells in small intestine tissue and mesenteric lymph node tissue.Immunohistochemical staining method was used to detect Casepase-1 protein in small intestine tissue and mesenteric lymph node tissue,as well as secretory im-munoglobulin A(sIgA)in small intestine mucosal tissue.Results There was a small amount of inflammatory cell in-filtration in the small intestine tissue of the zingerone group rats,but the degree of tissue congestion and edema was reduced compared to the model group.Compared with the model group,the levels of ET,IL-1β,TNF-α,NO,MPO and intesti-nal vascular permeability in the zingerone group were decreased(P<0.05).The number of NLRP3 and ASC positive cells in small intestine tissue and mesenteric lymph nodes,as well as the expression level of Casepase-1 protein in the zingerone group,were decreased(P<0.05).The expression level of sIgA protein in the small intestine mucosa in the zingerone group was increased(P<0.05).Conclusion Zingerone has a protective effect on the intestinal mucosal bar-rier of SAP rats,and zingerone has a protective effect on the intestinal mucosal barrier in SAP rats,which may be related to upregulated sIgA levels,inhibited NLRP3 inflammatory complex activation,and modulated intestinal immune function.关键词
