Effects of hsa_circ_100395 on proliferation,invasion and migration of HGC-27 and MGC-803 gastric cancer cells

Abstract
Aim To explore the effects of hsa_circ_100395 on proliferation,invasion and migration of gastric cancer cells.Methods The gastric cancer tissues and adjacent tissue samples were collected from 72 patients with gastric cancer,qRT-PCR was used to detect the expression of hsa_circ_100395 in gastric cancer tissues,and the relation-ship with clinicopathological characteristics was analyzed.The gastric cancer cell lines HGC-27 and MGC-803 were divid-ed into overexpression group(transfected with hsa_circ_100395 overexpression plasmid),silence group(transfected with hsa_circ_100395 silent plasmid)and control group(transfected with empty plasmid).CCK-8 method,scratch healing test,Transwell method,and nude mouse tumor formation test were used to detect the proliferation,migration and invasion ability of gastric cancer cells.Results The expression of hsa_circ_100395 was significantly down-regulated in gastric cancer.There were significant differences in tissue differentiation,infiltration depth,and TNM staging between low ex-pression and high expression of hsa_circ_100395(P<0.05).Compared with the control group,the overexpression of hsa_circ_100395 significantly inhibited the proliferation,migration,invasion of HGC-27 and MGC-803 cells and tumor growth,whereas hsa_circ_100395 silence significantly promoted the proliferation,migration,invasion of HGC-27 and MGC-803 cells and tumor growth.Conclusion Hsa_circ_100395 is expressed at low level in gastric cancer tissues,and overex-pression of hsa_circ_100395 can inhibit the proliferation,invasion and migration of gastric cancer cells.关键词
