Study on antagonistism of Lactobacillus salivarius against Escherichia coli based on intestinal mucosa morphology

Abstract
In order to investigate the antibacterial effect of Lactobacillus salivarius from chicken on Escherichia coli(ATCC25922),a strain of Lactobacillus salivarius was isolated and identified from the saliva of Xinjiang Black Chicken by selective medium combined with molecular biology.Oxford cup test was used to determine the antibacterial ability of Lactobacillus salivarius against Escherichia coli in vitro.Five week old SPF mice were randomly divided into blank control group(normal saline),inflammatory control group(1.5×108 CFU/mL),Lactobacillus salivarius group(1.5×108 CFU/mL)and antagonistic group(Lactobacillus salivarius + Escherichia coli).A mouse model of Escherichia coli infected by intragastric administration was established.The height of intestinal villus and the depth of crypt were observed and measured to evaluate the protective effect of Lactobacillus salivarius on the intestinal mucosa of mice and its ability to resist Escherichia coli.Vitro results showed that Lactobacillus salivarius from chicken had a good antibacterial effect on Escherichia coli.The results of histological sections showed that the Lactobacillus salivarius group had the longest villus height,the shallowest crypt depth,the highest villus/crypt ratio(P<0.01),and a large number of goblet cells arranged in order.The inflamma-tory control group had serious villus damage,disorderly arrangement of goblet cells,and increased crypt depth.The results showed that chicken Lactobacillus salivarius had a strong ability to antagonize Escherichia coli infection,could increase the velvet ratio of the intestinal mucosa,and had better protection and repair effects on the integrity and continuity of the intestinal mucosa in mice.The results of this study provide new ideas for the prevention and treatment of Escherichia coli infectious diseases.关键词
