Abstract
Objective:To explore the culture methods of different pathological types of cervical cancer organoids,and to conduct a preliminary evaluation of the successfully estab-lished organoid model.Methods:Fresh cervical cancer tissues were collected and used for or-ganoid culture and histological studies respectively.Organoid growth was observed using invert-ed microscope.HE staining was used to compare differences between cervical cancer organoids and orthotopic tumor tissues.Immunohistochemistry was carried out to evaluate the expression of tumor-associated proteins.Results:Different pathological types of cervical organoids were estab-lished successfully and the culture model was optimized.Inverted microscope revealed increased proliferation and continuous growth in volume of cervical cancer organoids.After 12-day cultiva-tion,the organoid volume reached a stable state.HE staining showed significant difference in morphologies between different pathological types of cervical cancer and confirmed the organoids had similar morphology to orthotopic tumor tissues.Immunohistochemically,the organoids stained positive for p16 and Ki67,similar to in situ tumor.Conclusion:This study preliminarily established an organoid culture method of human cervical cancer with different pathological types,which will provide a new model for basic research of cervical cancer and targeted drug screening.关键词
