青海汉族妊娠期高血压疾病Flt-1基因rs722503多态性分子特征分析
Molecular analysis of Flt-1 rs722503 polymorphism in hypertensive disorder of pregnan-cy in Han nationality of Qinghai

Abstract
Objective:To investigate the relationship between rs722503 polymorphism of fms-like tyrosine kinase-1(Flt-1)gene and hypertensive disorder of pregnancy(HDP)in Han nationality of Qinghai Province and to determine the expression of this gene in HDP group and control group preliminarily.Methods:A total of 143 Han nationality HDP subjects(HDP group)and 133 Han nationality normal pregnant subjects(control group)were selected from Qinghai province.The rs722503 typing in HDP group and control group was analyzed by poly-merase chain reaction-restriction fragment length polymorphism(PCR-RFLP)and verified by sequencing.The expression of Flt-1 mRNA in the placental tissues of the two groups were detec-ted by real-time PCR.Serum soluble fms-like tyrosine kinase-1(sFlt-1)levels of the two groups were detected by enzyme-linked immunosorbent assay(ELISA).Results:The frequency of AA,AG,GG genotype of rs722503 in HDP group and control group were 54.55%,41.26%,4.19%and 40.60%,50.38%,9.02%respectively(P＜0.05),there were significant differ-ences between the two genotypes,AA genotype(OR=1.756,95%CI=1.89～2.830,P＜0.05)was a risk factor for HDP.The frequency distribution of alleles A(OR=1.143,95%CI=1.024～1.275)and G(OR=0.726,95%CI=0.559～0.943)of rs722503 were significant different between HDP group and control group(P＜0.05).The relative expression of Flt-1mRNA in pla-cental tissue of HDP group was higher than that of control group(P＜0.01).The serum sFlt-1 level in HDP group[(7.04±1.94)ng/mL]was higher than that in control group[(2.92±1.02)ng/mL](P＜0.01).Conclusion:The rs722503 polymorphism of Flt-1 gene is signifi-cantly correlated with HDP of Han nationality in Qinghai Province,the AA genotype may be the susceptibility genotype of HDP,the A allele may be the susceptibility gene of HDP,the G allele may be the protective gene of HDP.The relative expression of Flt-1mRNA in placental tissue and the level of serum sFlt-1 protein may be correlated with HDP of Han nationality in Qinghai Province.关键词
