除虫脲(DFB)属于苯甲酰苯脲类化合物,能够抑制昆虫及其幼虫的几丁质合成,从而控制成虫的数量.为了解除虫脲预混剂混饲给药后,猪粪便中药物的浓度,以DFB-13C6为内标,建立了猪粪便中DFB药物浓度的液相色谱-串联质谱(LC-MS/MS)检测方法.以冰乙酸-乙腈提取粪便中的DFB,采用QuEChERS技术净化提取液,LC-MS/MS测定,内标法定量.结果显示,猪粪便样品基质对于DFB的定量检测干扰较小,检测方法灵敏度高,检测限为 10 ng/g,定量限为 20 ng/g;检测方法的基质效应对样品的检测影响较小;标准溶液在 2~500 ng/mL的浓度范围内,线性关系良好(R2≥0.99);在定量限20 ng/g及低浓度(100 ng/g)、中浓度(1000 ng/g)、高浓度(5000 ng/g)的添加水平下,平均准确度在93%~110%,批内、批间变异系数均小于 10%,符合方法学要求;不同浓度加标样品处理后的溶液,室温放置 24h稳定性良好,但应避免样品的反复冻融.结果表明,本试验建立的LC-MS/MS检测方法可用于猪粪便中DFB药物浓度的检测,为后期开展除虫脲预混剂的临床试验提供了可靠方法.

Diflubenzuron(DFB)belongs to benzoylphenyl ureas compounds,which inhibit the chitin synthesis and development of insect and their larvae.In order to investigate the concentration of DFB in swine feces after administration of diflubenzuron premix through mixed feeding,a method of liquid chromatography-tandem mass spectrometry(LC-MS/MS)with DFB-13 C6 as the internal standard(IS)was de-veloped to determine DFB in the feces.The DFB in swine feces samples was extracted by glacial acetic acid-acetonitrile,and the extracted solution was cleaned up by QuEChERS technology.The DFB was determined by LC-MS/MS and quantified using the internal standard meth-od.The results showed that the matrix of swine feces had little interference with the quantitative detection of DFB,and the method had high sensitivity with a limit of detection of 10 ng/g and a limit of quantitation(LOQ)of 20 ng/g.The matrix effect of the method had little influ-ence on the detection of the samples.The linearity was good(R2≥0.99)when the standard solutions were in the concentration range of 2 ng/mL to 500 ng/mL.The average accuracy of the method was between 93% and 110% and the intra-assay and inter-assay precisions were both less than 10% when the spiked concentrations were at LOQ 20 ng/g,and at low(100 ng/g),medium(1000 ng/g)and high(5000 ng/g)added concentrations.These results met the methodological requirements.The processed solution at different spiked concentrations had good stability when placed at room temperature for 24 h.However,repeated freeze-thaw of the sample should be avoided.The proposed method might be employed to determine DFB concentration in swine feces,which would provide a reliable method for future clinical trials of diflubenzuron premix.
