Abstract
Objective To analyze the mechanism of catclaw buttercup root tuber in the treatment of breast cancer based on network pharmacology and molecular docking.Methods The eligible active ingredients and target proteins were obtained through the TCMSP database and converted into target genes using the UniProt database.The GeneCards database was used to search for the target of"breast cancer".The target genes of the two were intersected by Venny software,and the network diagram was constructed by Cytoscape 3.9.1 software.PPI network was constructed based on STRING database and imported into Cytoscape 3.9.1 software for analysis.GO function and KEGG pathway enrichment analysis of drug targets were performed using DAVID database.Results A total of 10 active components of catclaw buttercup root tuber were found to be effective in breast cancer,of which Truflex OBP,Qtvittadinoside_qt and Mandenol were the core components.There were 26 common targets,including CASP8,BAX,PGR,BCL2,PTGS2 and other genes,and their signaling pathways were mainly concentrated in PI3K-AKT and VEGF signaling pathways.Molecular docking showed that Sitosteryl acetate had the lowest affinity with PRKCA,BAX and CASP8,and the binding ability was strong;Mandenol had the lowest affinity with CASP9 and the strongest binding ability.Conclusion The mechanism of catclaw buttercup root tuber on breast cancer may be through multi-component,multi-target and multi-signal pathway.关键词
