Effect of ZBD-2 on improving fear-,anxiety-and depression-like behaviors in mice model of posttraumatic stress disorder

Abstract
Objective To investigate the effect of translocator protein 18 kDa(TSPO)ligand ZBD-2 on the behaviors of mice model of posttraumatic stress disorder(PTSD).Methods Forty-eight male C57BL/6 mice were randomly divided into normal control(Control)group,PTSD model(Model)group,PTSD model+ZBD-2(low,medium and high dose)group,and PTSD model+paroxetine positive control(Model+Paro)group,with 8 mice in each group.A PTSD mouse model was established by using conditioned fear(CF)combined with single-prolonged stress.The effect of ZBD-2 on fear memory and the improvement of anxiety-and depression-like behaviors in PTSD model mice were observed by behavioral open field test,elevated plus maze test,tail suspension test and fear memory test.Results Compared with the Control group,the total distance and time of movement in the central area of open field were significantly shortened(P＜0.01),the number of times and retention time of entering the open arms of the elevated plus maze were significantly reduced(P＜0.01),the suspension time was prolonged(P＜0.01),and the duration of immobilization in CF was significantly prolonged(P＜0.01)in the Model group,indicating that the PTSD model was successfully prepared.Compared with the Model group,the PTSD model+ZBD-2 group and the Model+Paro group significantly improved the aforementioned behaviors(P＜0.05,P＜0.01),and the PTSD model+ZBD-2 group had the best effect at meldium dose.Conclusion The TSPO ligand ZBD-2 can effectively improve fear-,anxiety-and depression-like behaviors in PTSD model mice,and it can be used as a potential candidate drug for the treatment of PTSD.关键词
