Abstract

Objective To design and synthesize a novel dihydroxanthene(DHX)structure near-infrared fluorescence probe for in situ,trace,visualization,and specific detection of hydrogen peroxide(H2 O2).Methods A novel mitochondrial targeted fluorescence probe molecule DHX-H2O2 was constructed with DHX structure as fluorophore and pyridine salt as acceptor.The fluorescence properties,selectivity,relationship with H2O2 concentration and recognition mechanism of the probe were studied.In addition,the application of the probe in the detection of H2O2 in cells and zebrafish were also studied.Results The probe DHX-H2 O2 could achieve near-infrared(λem=675 nm)fluorescence response to H2O2 with good selectivity and excellent sensitivity(4.21 μmol/L).In addition,the probe had the ability to locate the mitochondria of cells,with a Pearson correlation coefficient of 0.97.Conclusion The designed and synthesized probe molecule has the advantages of good selectivity,high sensitivity and low cytotoxicity,and has successfully realized the imaging detection of endogenous and exogenous H2O2 in cells and zebrafish,which has broad biological application prospects.
