Identification of Tick Species of Stray Cat Origin and PCR Detection for Rickettsia Carried by Them in Alar City,Xinjiang Uygur Autonomous Region

Abstract
In order to investigate tick species on the body surface of cats and Rickettsia carried by them,tick samples were collected from stray cats in a residential area of Alar,Xinjiang to identify tick species using the morphological and molecular biology method,and to detect the Rickettsia carried by ticks using PCR.Then the genotype of Rickettsia was identified through sequence matching,and a genetic evolution tree was constructed to analyze its molecular genetic characteristics.The results revealed that one of the 4 stray cats had 3 ticks parasitized on its body surface,which were preliminarily identified as Rhipicephalus turanicus through morphological observation;based on the mitochondrial SSU rRNA and COX Ⅰ genes,the DNA samples of the three ticks were amplified by PCR,and the ticks were finally identified as Rhipicephalus turanicus.Based on OmpA and OmpB genes of Rickettsia,the DNA samples of the three ticks were amplified by PCR and their sequences were compared and analyzed,it was found that the Rickettsia sequence detected in the DNA of two ticks was 100%homologous to the Candidatus R.barbariae from Rhipicephalus turanicus in China and Rhipicephalus annulatus in Lebanon,hence the Rickettsia was identified as Candidatus R.barbariae.In conclusion,there were Rhipicephalus turanicus parasitized on the body surface of stray cats in Alar,which could carry Candidatus R.barbariae.A basic data was thus provided for researches on parasitic ticks of cat origin and their pathogen carrying status in China.关键词
