Selection of Production Strains for Trivalent Inactivated Vaccine against Infectious Coryza

Abstract
In order to select strains with good immunogenicity for production of inactivated vaccines against avian infectious coryza,this study analyzed the pathogenicity and immunogenicity of 5 strains of serotype A,3 strains of serotype B,and 3 strains of serotype C Avibacterium paragallinarum.Initially,strains with strong virulence were selected through virulence test,and the minimum pathogenic dose of the selected strains was determined.This dose was then set as the challenge dose for subsequent immunogenicity analysis and minimum antigen content determination.The results of virulence test showed that all 11 isolated strains exhibited strong virulence.Among them,2 strains of serotype A(HN3 and HB2),2 strains of serotype B(HN5 and SD4),and 2 strains of serotype C(HN1 and SD3)with strong virulence were selected.The results of the minimum pathogenic dose determination showed that serotype A strain HN3,serotype B strain HN5,and serotype C strain SD3 had the strongest toxicity,with minimum pathogenic doses of approximately 2.0 × 104,1.8 × 103,and 2.3 × 103 CFU,respectively.The results of immunogenicity analysis and minimum antigen content determination showed that serotype A strain HN3,serotype B strain HN5,and serotype C strain SD3 had excellent immunogenicity.These strains not only resisted attacks from homologous strains but also provided good protection against heterologous strains of the same serotype.The post-immunization serum HI antibody titers and positive rates were high,and there was a good correlation with protection against challenge.The minimum antigen contents for serotype A strain HN3,serotype B strain HN5,and serotype C strain SD3 was determined to be 5.0 × 108,1.0 × 109,and 1.5 × 109 CFU/mL,respectively.Therefore,serotype A strain HN3,serotype B strain HN5,and serotype C strain SD3 of Avibacterium paragallinarum,which exhibit strong virulence and good immunogenicity,can be used as production strains for a trivalent inactivated vaccine against avian infectious coryza.关键词
