鲤鱼双线绦虫裂头蚴的分子鉴定和系统发育关系分析
Molecular Identification and Phylogenetic Relationships Analysis of Digramma interuota in Cyprinus carpio

Abstract
To identify the"noodle-like"worm parasites found in the abdominal cavities of common carp from a fish farm in Tianjin,PCR was used to amplify the 18S rRNA,CO Ⅰ,and COB gene sequences of the worms.Molecular identification and gene sequencing were performed,and the homology,molecular evolution,and phylogenetic relationships based on these three genes were analyzed.The results indicated that the 18S rRNA,CO Ⅰ,and COB gene sequences of the worms parasitizing the Cyprinus carpio showed high homology with nucleotide sequences from Digramma interuota isolates MN204040.1(China),MN219466.1(China),and EU241209.1(France),with homologies of 99.45%,98.74%,and 99.26%,respectively.Phylogenetic trees constructed based on the 18S rRNA,CO Ⅰ,and COB gene sequences showed that the tapeworm plerocercoid isolated in this study clustered in the same branch as the Digramma interuota species identified in the GenBank database,and they were closely related to the Digramma interuota isolate from Wuhan,China.This study provides initial insights into the phylogenetic relationship of the Digramma interuota isolate obtained in Tianjin with other species,supporting molecular identification and phylogenetic analysis of Digramma interuota.关键词
