摘要

Abstract

This study aims to reveal the spread and drug resistance of Salmonella in the process of pork production by monitoring the contamination distribution and drug resistance of Salmonella in each link of pig slaughter and pork products.Five representative pig slaughterhouses in Shandong province were selected as risk monitoring sites.A total of 552 samples,including anal swabs,environmental swabs,water samples,and carcass swabs,were collected.Salmonella isolates were isolated and identified using the national food safety standards and Matrix-assisted laser desorption ionization time-of-flight mass spectrometry(MALDI-TOF MS).The drug sensitivity of Salmonella isolates was determined using the microbroth dilution method.Multilocus sequence typing(MLST)was used to identify the genotypes of Salmonella.The results showed that a total of 58 Salmonella strains were isolated,with a total positive detection rate of 10.5%(58/552).Salmonella contamination was more severe in small and medium-scale slaughterhouses(15.6%,33/212)than in large-scale slaughterhouses(7.4%,25/340).The highest detection rate was observed in the samples collected in pre-cooling stage of the slaughtering process,with a rate of 26.7%(8/30).Among different types of samples,the highest detection rate was in the anal swab samples,with a rate of 15.1%(16/106).The highest drug resistance rate was observed for ampicillin(96.6%,56/58),and the multi-drug resistance rate was 96.6%(56/58).Salmonella isolates from small and medium-scale slaughterhouses exhibited more severe overall drug resistance.The multi-drug resistance rate was higher in carcass swabs(46.4%,26/56)and anal swab samples(28.6%,16/56)than in environmental swab samples(25.0%,14/56).MLST gene typing revealed that the 58 Salmonella isolates corresponded to 6 ST types,with ST40 being the most widespread and detected in all stages,followed by ST19.The results indicate that Salmonella contamination is severe during the pig slaughtering process,highlighting the need to strengthen hygiene control during slaughter.Pig slaughterhouses of all scales should pay attention to the transmission of drug-resistant bacteria and genes in the industry and food chains,and develop reasonable prevention and control measures to prevent potential food safety risks.
