Abstract
Objective To evaluate the efficacy and safety of Paliperidone Palmitate Long-Acting Injection(PP-LAI)and Risperidone Long-acting Injection(RLAI)for schizophrenia.Methods Databases including CNKI,VIP,PubMed,and The Cochrane Library were searched to collect Randomized Controlled Trials(RCTs)on PP-LAI and RLAI.The Cochrane Collaboration's tool was used to access the risk of bias,and RevMan 5.4 was used for meta-analysis,to compare the differences between the two drugs.Results 10 RCTs were eventually included.There are 10 efficacy analysis sets involving 2 629 patients and 9 safety analysis sets involving 2 984 patients.The results of the meta-analysis showed that there are no significant differences between PP-LAI and RLAI in effective rate,change from baseline to endpoint in PANSS and PSP score,the incidence of all adverse events(AEs),cardiac-related AEs,sleep disorders and prolactin-related AEs(P＞0.05).However,the incidence of Extra Pyramidal Symptoms(EPS)and glucose-related AEs caused by PP-LAI was significantly lower than that caused by RLAI(P＜0.05).Conclusion PP-LAI and RLAI are two long-acting antipsychotic agents commonly used in clinics.Their efficacy and safety in the treatment of schizophrenia are roughly equal,but the incidence of EPS and glucose-related AEs caused by PP-LAI is lower.Clinical medication should be selected individually based on patients'symptoms,medical history and economic level.关键词
