摘要

Abstract

Objective To investigate the expression,proliferation,molecular signaling pathway and immune microenvironment of CCT3(Chaperon in containing TCP 1,subunit 3)in papillary thyroid carcinoma.Methods The expression data and clinical data of thyroid cancer downloaded from TCGA database were used to analyze the difference of CCT3 expression between thyroid cancer and adjacent tissues.30 cases of papillary thyroid carcinoma and adjacent tissues were collected,and the expression of CCT3 protein was detected and verified by immunohistochemical staining.RNA interference technology was used to silence the expression of CCT3 gene in thyroid papillary carcinoma cell line(K1)as interference group,while CCT3 was highly expressed as control group;RT-qPCR and Western Blot were used to detect the silencing efficiency,MTT and flow cytometry were used to detect the effects of cell proliferation,cell cycle and apoptosis in the two groups.GSEA enrichment analysis was used to predict the signal pathway of CCT3 in thyroid carcinoma,and CIBERSORT method was used to analyze the correlation between CCT3 expression and immune infiltration.Results The results of TCGA database showed that CCT3 gene in papillary thyroid carcinoma was higher than that in normal tissues adjacent to the carcinoma(P<0.01).The expression of CCT3 protein and mRNA was up-regulated in clinical samples and in vitro experimental results(P<0.001).Compared with the control group,silencing CCT3 can significantly inhibit the proliferation of papillary thyroid cancer cells,increase the apoptosis,and arrest the cell cycle in G2/M phase(P<0.05).GSEA enrichment analysis and CIBERSORT analysis showed that CCT3 was mainly enriched in immune-related pathways and acted on the occurrence and development of TC through memory B cells,plasma cells,activated memory CD4+T cells,macrophage M1,macrophage M2,resting mast cells and eosinophils.Conclusions CCT3 is closely related to the occurrence and development of papillary thyroid carcinoma and CCT3 may become a new molecular marker for diagnosis,diagnosis and treatment of papillary thyroid carcinoma.
