豚鼠被动皮肤过敏试验条件研究
Study on the Conditions of Passive Cutaneous Anaphylaxis Test in Guinea Pigs

Abstract
Objective To investigate the optimal conditions(allergenic positive substance and time)of passive cutaneous anaphylaxis(PCA)test in guinea pigs.Methods Ovalbumin(ova,0.4%)and bovine serum albumin(BSA,1%)were used as the PCA antigens,the PCA test was performed on guinea pigs from different manufacturers(Weifang Shengnuo Experimental Animal Breeding Co.,Ltd.,Qingdao Kangda Biological Technology Co.,Ltd.,Jinan Jinfeng Experimental Animal Co.,Ltd.,hereinafter referred to as the Shengnuo,Kangda and Jinfeng respectively)at different sensitization time(4 h and 24 h),and the diameter of the blue spots was recorded.Results After the passive sensitization with ova for 4 h,the blue spots of guinea pigs from three manufacturers had clear boundaries,and the diameters were greater than 5.0 mm(positive);while after the passive sensitization with ova for 24 h,the above blue spots were mixed,with unclear boundaries and unable to be measured.After passive sensitization with BSA for 4 h,the blue spots of guinea pigs from Shengnuo and Kangda had clear boundaries,and the diameters were greater than 5.0 mm,while the blue spots of guinea pigs from Jinfeng sensitized by only serum containing antibodies(1∶2,1∶4,V/V)had a diameter greater than 5.0 mm.After passive sensitization with BSA for 24 h,the blue spots of guinea pigs from Jinfeng had clear boundaries,and the diameters were greater than 5.0 mm,while the blue spots of guinea pigs from Shengnuo and Kangda were mixed and indistinguishable,which were unable to be measured.Conclusion In PCA test of guinea pigs,0.4%ova solution is preferred as the test antigen,and the sensitization time of 4 h is suitable.关键词
