海南产温郁金悬浮细胞培养及其挥发性成分的积累研究
Study on Suspension Cell Culture and Volatile Components Accumulation of Curcuma wenyujin in Hainan

Abstract
Objective:To establish a suspension cell line of Curcuma wenyujin introduced from Hainan,and to study the accumula-tion of volatile components in culture materials in vitro.Methods:The callus-inducing explants,culture-medium and suspension cell initi-ation callus were selected to establish stable suspension cell lines.The volatile components in culture materials in vitro such as suspen-ded cell mass of Curcuma wenyujin were determined by headspace solid phase microextraction-gas chromatography-Mass spectrometry.Results:The callus induced by young roots was a suitable starting material for the establishment of stable suspension cell lines.After secondary culture for two times,the callus proliferated obviously,and it was easy to form embryogenic callus with bright yellow color,u-niform granules,loose and fragile.Among the three culture materials in vitro,the detection rates of total volatile substances were tissue culture seedlings,suspended cell mass,callus in order.The detection rates of common chemical ingredients of Curcuma wenyujin volatile oil,including germacrone,curzerene,curdione,elemene,were tissue culture seedlings,callus,suspended cell mass in order.Conclusion:A certain amount of volatile active components such as zedoary oil are accumulated in the culture materials in vitro such as the suspen-ded cell line of Curcuma wenyujin introduced in Hainan,which provides basic research for further screening and optimization of culture system and promoting the accumulation of secondary metabolites in culture.关键词
