Abstract
Objective:To establish an effective method for the identification of Buthus martensii and its adulterants Lychas mucr-onatus.Methods:Based on the mitochondrial cytochromic C oxidase subunit(CO Ⅰ)gene differences between Buthus martensii and Ly-chas mucronatus,specific primers were designed to distinguish Buthus martensii and Lychas mucronatus.The multiple PCR methods were established,the annealing temperature,primer concentration,cycle number and amount of DNA template were optimized and screened.The Buthus martensii and Lychas mucronatus from different sources were determined,and the specific band size was used for identifica-tion.Results:When the annealing temperature was 51.6 ℃,the number of cycles was 30,and the amount of DNA template was 30 ng,the specific bands of about 342 bp and 108 bp appeared in Buthus martensii and Lychas mucronatus,respectively.Conclusion:This method can be used for rapid and accurate identification of Buthus martensii,Lychas mucronatus and doping samples,and can make up for the shortcomings of traditional identification methods.关键词
