Identification of DNA Barcoding of Corydalis Plants in Tibet

Abstract
Objective:To use ITS2 sequence and psbA-trnH sequence as DNA barcodes for molecular identification of Corydalis plants from Tibet.Methods:By comparing the amplification success rate and sequencing success rate of the two sequences,the sequence characteristics were analyzed,the intraspecic and interspecific genetic distances were calculated.Neighbor-joining evolutionary tree was constructed to evaluate the identification efficiency of the two sequences.Results:The success rate of amplification and sequencing of the two sequences was 100%.The maximum intraspecific genetic distance of psbA-trnH sequence was smaller than the minimum inter-specific genetic distance,which met the genetic distance requirement of DNA barcodes.In phylogenetic tree analysis,psbA-trnH se-quence could effectively distinguish different Corydalis plants,but ITS2 sequence had poor clustering effect.Conclusion:psbA-trnH se-quence has high identification efficiency and can be used as DNA barcodes for the identification of Corydalis plants in Tibet.The study lays a solid foundation for the further study on the genomics of Corydalis plants in Tibet and provides a new method for the quality con-trol of Tibetan medicine of Corydalis plants in Tibet.关键词
