摘要

Abstract

Objective:To establish UHPLC-MS/MS method for simultaneous determination of 16 saponins in Notoginseng Radix et Rhizoma(ginsenosides Rf,Rg1,Rg2,Re,Rd,Rb1,Rb2,Rb3,Rc,Fa,Rk1,Rg5,Rg3,F2 and notoginseng saponins R1,Fc)in rat plasma,and to investigate their pharmacokinetic characteristics.Methods:The plasma samples were treated with methanol-acetonitrile(4:1)pre-cipitated protein,and ACQUITY UPLC BEH C18(100 mmx2.1 mm,1.7 μm)column was used.The mobile phase was 0.1%formic acid-acetonitrile with gradient elution;Using electrospray ion source(ESI),multiple response monitoring(MRM)was used to detect negative ions,DAS 3.0 software was used to calculate the pharmacokinetic parameters.Results:The 16 saponins had a good linear rela-tionship with the peak area in the corresponding concentration range(r>0.9900).The RSD of intra-day and inter-day precision ranged from 0.88%to 14.41%,and accuracy ranged from 89.60%to 114.00%.The extraction recovery was 84.67%to 111.87%,and the matrix effect was 77.04%to 117.64%.Conclusion:The established UHPLC-MS/MS analytical method in the study is accurate and sen-sitive,and can be used for the content determination and pharmacokinetic study of the 16 saponins in Notoginseng Radix et Rhizoma in rats.
