消积化肝方调节Wnt/β-catenin对肝癌细胞恶性生物学行为的影响
Effect of Xiaoji Huagan Recipe on Malignant Biological Behavior of Hepatoma Cells by Regulating Wnt/β-catenin
摘要
Abstract
Objective:To investigate the influence of Xiaoji huagan recipe on the malignant biological behavior of liver cancer cells by regulating Wnt/β-catenin.Methods:HepG2 cells in logarithmic growth phase were divided into control group(20％blank serum),low dose of Xiaoji huagan recipe containing serum(10％)group,medium dose of Xiaoji huagan recipe containing serum(20％)group,high dose of Xiaoji huagan recipe containing serum(30％)group,LiCl(10 mmol/L)group,high dose of Xiaoji huagan recipe containing se-rum with LiCl(30％Xiaoji huagan recipe containing serum with 10 mmol/L LiCl)group,the proliferation of HepG2 cells was detected by CCK-8 and EdU staining.The apoptosis of HepG2 cells was detected by flow cytometry.The migration of HepG2 cells was detected by scratch test.Transwell was used to detect the HepG2 cell invasion.Western Blot was used to detect the expressions of PCNA,Bax,MMP-2,MMP-9,Wnt3a,β-catenin in HepG2 cells.Results:Compared with the control group,the D(λ)450 value,EdU positive rate,scratch healing rate,number of cell invasion and PCNA,MMP-2,MMP-9,Wnt3a,β-catenin protein expressions in the each dose of Xiaoji hua-gan recipe containing serum group were significantly decreased,the cell apoptosis rate and Bax protein expression were significantly in-creased,D(λ)450 value,EdU positive rate,scar healing rate,number of cell invasion and PCNA,MMP-2,MMP-9,Wnt3a,β-catenin pro-tein expressions were significantly increased in LiCl group,while the cell apoptosis rate and Bax protein expression were significantly de-creased(P＜0.05).Compared with high dose of Xiaoji huagan recipe containing serum group,D(λ)450 value,EdU positive rate,scratch healing rate,number of cell invasion and PCNA,MMP-2,MMP-9,Wnt3a,β-catenin protein expressions in HepG2 cell of high dose of Xiaoji huagan recipe containing serum with LiCl group were significantly increased,apoptosis rate and Bax protein expression were sig-nificantly decreased(P＜0.05).Conclusion:Xiaoji huagan recipe can inhibit the proliferation,migration and invasion of HepG2 cells and promote apoptosis by inhibiting Wnt/β-catenin pathway.关键词
