基于HPLC指纹图谱的沙棘叶黄酮类功效成分研究
Study on Flavonoids from Seabuckthorn Leaves Based on HPLC Fingerprint

Abstract
Objective:To comprehensively evaluate the quality of seabuckthorn leaves from different producing areas,the HPLC fingerprint was established and the contents of isorhamnetin,quercetin and kaempferol were also determined.Methods:The analysis of fingerprints was performed on a C18 column(250 mm×4.6 mm,5 μm).The mobile phase consisted of acetonitrile:0.4%phosphoric acid aqueous solution with gradient elution.The column temperature was 30℃and the flow rate was 0.8 mL·min-1.The injection volume was 10 μL and the detection wavelength was 280 nm.The similarity evaluation of fingerprint was carried out by the software of"Similarity Evaluation System for Chromatographic Fingerprint of TCMs"(2012 version).The content determination was performed on a C18 column(250 mm×4.6 mm,5 μm).The mobile phase consisted of methanol:0.1%formic acid aqueous solution(58:42)at the flow rate of 1.0 mL·min-1.The column temperature was 30℃.The injection volume was 10 μL and the detection wavelength was 370 nm.Results:HPLC fingerprints of seabuckthorn leaves from different producing areas were established.The similarity of leaves from different producing areas was 0.647-0.910.There were 26 common peaks and 9 common peaks were identified as pedunculagin,hippophaenin B,casuarinin,epicatechin,narcissoside,epigallocatechin,rutin,isoquercitrin,and kaempfer-3-O-rutinoside,by comparison with the reference substance.In addition,the contents of isorhamnetin,quercetin and kaempferol were quantified by HPLC,the results showed that the content of quercetin,kaempferol and isorhamnetin were 0.12%-0.33%,0.07%-0.32%and 0.10%-0.49%,respectively.The highest content of quercetin was 0.33%in Xinzhou,Shanxi.The highest content of kaempferol was 0.32%in Ordos,Inner Mongolia Autonomous Region.The highest isorhamnetin content was 0.49%in Tacheng regiment of Xinjiang.Conclusion:The method used in this study has good precision,stability and reproducibility,which can effectively evaluate the quality of seabuckthorn leaves.The contents of flavonoids in seabuckthorn leaves of different producing areas have significant differences.关键词
